ABSTRACT

ObjectiveTo investigate the effect of Jiawei Bazhen Yimu capsule on serum sex hormones, sexual organs and estrogen signaling pathway in female rats with premature ovarian failure. MethodThe key target proteins of Jiawei Bazhen Yimu capsule in the treatment of premature ovarian failure were screened out by network pharmacology analysis. Female healthy SD rats were selected, and the rat model of premature ovarian failure was established by ovariectomy. Fifty ovariectomized rats were randomly divided into a model group, an estradiol (E2) valerate group, and Jiawei Bazhen Yimu capsule low, medium, and high-dose groups. Another 10 healthy female rats were set as a sham operation group. The sham operation group and the model group were given distilled water by gavage, and other administration groups were given corresponding doses of drugs by gavage. After 21 d, the serum hormone levels of female rats were measured, including E2, progesterone (P), follicle stimulating hormone (FSH), and luteinizing hormone (LH). Immunofluorescence staining (IF) was used to detect the protein expression levels of estrogen receptor 1 (ESR1), estrogen metabolism P4503A4 enzyme (CYP3A4), and P45019A1 enzyme (CYP19A1) in the uterine tissues of female rats. ResultAs compared with the model group, the serum E2 and P levels of female rats in the Jiawei Bazhen Yimu capsule low, medium, and high-dose groups were significantly increased. Jiawei Bazhen Yimu capsule improved the endometrial status of female rats and increased positive expression of ESR1, CYP3A4, and CYP19A1 in the uterine tissues of female rats (P<0.05). ConclusionThe mechanism of Jiawei Bazhenyimu capsule in the treatment of premature ovarian failure may be related to its hormone-like effect and activation of the estrogen signaling pathway.

ABSTRACT

Objective:To observe the effect of combining transcranial direct current stimulation (tDCS) with functional electrical stimulation-assisted cycling (FES-cycling) on lower limb motor function early after a stroke.Methods:Thirty-seven survivors of a recent stroke were divided into a tDCS treatment group ( n=18) and a pseudo-stimulation group ( n=19). While receiving routine rehabilitation training and clinical drug treatment, the tDCS treatment group also cycled in response to functional electrical stimulation while simultaneously receiving tDCS anode stimulation of the motor cortex M1 area. The pseudo-stimulation group followed the same protocol but with the tDCS stimulation inactivated. Both groups were treated for 20min daily, 5 days weekly for 4 weeks. Before and after the 4 weeks of treatment, the lower limb motor function, walking ability and ability in the activities of daily living of both groups were evaluated using the Fugl-Meyer assessment scale for the lower extremities (FMA-LE), the timed up and go test (TUGT) and the modified Barthel index (MBI) respectively. Transcranial magnetic stimulation was used to detect each subject′s cerebral cortex motor threshold (CMT) , cortical latency (CL) and central motor conduction time (CMCT) as well as the amplitude (Amp) of the motor evoked potential of the lower limb primary motor cortex (M1 area). Results:After 4 weeks of treatment, the average FMA-LE and MBI scores and TUGT times of the two groups had improved significantly compared with those before treatment. The average FMA-LE score and TUGT time of the tDCS group were significantly better than those of the pseudo-stimulation group. The average CMT, CL and CMCT in both groups were significantly lower than those before the intervention, while the average Amp had increased significantly, but there were significant differences in the average CMT, Amp, CL and CMCT between the two groups after the 4 weeks of treatment. Conclusions:Transcranial direct current stimulation combined with cycling assisted by functional electrical stimulation can effectively stimulate excitability in the motor cortex soon after a stroke. That should promote the recovery of nerve activity and lower limb function.

ABSTRACT

Objective To investigate the impact of spatial distribution of emphysema on pulmonary function and clinical severity in patients with chronic obstructive pulmonary disease (COPD).Methods Chest MSCT was performed in 84 patients with COPD.The percentage of low attenuation areas were automatically calculated for each lobe with the COPD analysis software. The correlations between low-attenuation area percent(LAA%) of each lobe and the parameters of pulmonary function were analyzed.According to the pulmonary emphysema heterogeneity index, the patients were divided into non-emphysema group and emphysema group, and the latter were divided into the predominantly upper lobe subgroup and the predominantly lower-lobe subgroup. The differences of pulmonary function parameters between the 2 groups and clinical severity between the 2 subgroups in emphysema group were compared. Results LAA% of each lobe was negatively correlated with single breath carbon monoxide diffusing capacity to predicted value ratio (DLcoSB%pred).Except for the right middle lobe,LAA% of the remaining lobes were negatively correlated with forced expiratory volume in one second/forced vital capacity (FEV1/FVC) and FEV1%pred,while positively correlated with resonance frequency (Fres). The differences of pulmonary function parameters between the 2 groups were statistically significant (all P < 0.05).The differences of FEV1%pred and FEV1/FVC between the 2 subgroups in emphysema group were statistically significant (all P <0.05),and the differences of Fres and DLcoSB%pred between the 2 subgroups were not statistically significant (all P>0.05).The difference of clinical severity between the 2 subgroups was statistically significant (x2 =4.17,P=0.041).Conclusion The spatial distribution of pulmonary emphysema affects pulmonary function and clinical severity in COPD patients. The patients with the predominantly lower-lobe emphysema are relatively worse in lung function, and more serious in the clinical severity.

ABSTRACT

Objective To evaluate the applicability of Han strain bacterial artificial chromosome (Han-BAC) in small fragment mutation of the human cytomegalovirus (HCMV) genome. Methods A 31-bp long fragment of LUNA between UL80 and UL82 in the HCMV was chosen as the mutation target. Kanamycin resistance gene sequence flanking the homologous arms of the target neighbor sequence was used to replace the target sequence. Electronic transformation was used to rescue the mutated virus. Reverse transcription PCR and cDNA clone sequencing were used to identify the mRNA expression and the 3' terminal structures of LUNA,UL80,and UL82 transcripts. Results The 31 bp fragment was replaced precisely by the kanamycin resistance gene sequence. The efficiency of mutation was more than 3%. LUNA-mutated virus was rescued successfully. Transcriptions and 3' terminal structures of the UL80 and UL82 transcripts of the mutant virus were the same as those of its original virus. Conclusion The sequence at the transcription start site of LUNA was replaced successfully. The HCMV Han-BAC supports fragment mutation as small as 31 bp with a relatively high efficiency.

ABSTRACT

Objective To investigate the association between non-Hodgkin lymphoma (NHL) and hepatitis B virus (HBV). Methods The serum HBV markers in 305 NHL patients who were diagnosed in the Affiliated Hospital of Southwest Medical University from January 2014 to December 2016 was detected by automatic chemiluminescence immunoassay. The infection rate of HBV in NHL patients was compared with that in 312 colorectal cancer patients and the national general population (81775 peoples). Results The positive rates of hepatitis B surface antigen (HBsAg), hepatitis B surface antibody (HBsAb) and hepatitis B core antibody (HBcAb) in 305 patients of NHL were compared with the general population [19.0 % (58/305) vs. 7.2 % (5888/81775), 44.3 % (135/305) vs. 50.1 % (40969/81775), 45.9 % (140/305) vs. 34.1 %(27885/81775)], and the differences were statistically significant (χ2 values were 63.1, 4.1, 18.8, all P<0.05). The positive rate of HBsAg in NHL patients was compared with colorectal cancer patients and the general population, and the differences were statistically significant (χ2= 65.7, P< 0.01). The positive rate of HBsAg in B-cell NHL was statistically different from T-cell NHL [21.3%(51/239) vs. 10.6%(7/66),χ2=3.869, P<0.05]. But the positive rate of HBcAb and HBsAb in B-cell NHL were compared with T-cell NHL, and there was no statistically significant difference (both P> 0.05). Among 133 NHL patients, the HBV DNA positive rate was 33.1 % (44/133), and 74.1 % (43/58) in 58 cases of HBsAg-positive NHL, while 4.2 % (1/24) in 24 cases of HBsAg-negative but HBcAb-positive NHL. Conclusions The infection rate of HBV in NHL patients is higher than that in colorectal cancer patients and the general population, in which the occult HBV infection is worthy of much attention. The positive rate of HBsAg in T-cell NHL patients is lower than that in B-cell NHL patients. For NHL patients with HBV infection, anti-HBV treatment to prevent reactivation of the virus should be given before the anti-tumor treatment.

ABSTRACT

Objective To evaluate the effect of the transcranial magnetic stimulation on upper-extremity function rehabilitation and changes in the excitability of cerebral cortex, and to evaluate from the viewpoint of electro-physiology the prognosis so as to guide the rehabilitation treatment of patients after stroke. Methods Forty-six patients in the early stage after a stroke were given TMS examinations of the ipsilateral brain region. Those with the motor evoked potentials (MEPs) amplitudes lower than 50 μV were classified into a motion-induced experimental negative group (the negative group),whiie those whose MEP amplitude reached 50 μV or more were classified as movement-induced positive (the positive group). Both groups were given the same treatment. Before and after 2,4 and 8 weeks of treatment the Fugl-Meyer movement function rating scale was used to assess their bilateral upper limb movement function.TMS technology was used to detect any change in the resting motor threshold (RMT) and the amplitude (Amp) of MEPs in the motor cortex. The incubation period of the cortex (CL) and the central motor conduction time (CMCT) in the contralateral motor cerebral cortex were also observed. Results After 4 weeks of treatment, the average score of the positive group on Fugl-Meyer upper movement function rating scale reached (54.99±2.76),significantly higher than before treatment and significantly higher than the negative group's average (P<0.05).After 8 weeks of treatment, the average score in the positive group had increased further to 73.11±2.98,still significantly higher than that of the negative group (P<0.01).After treatment,RMT decreased progressively in both groups, but that of the negative group dropped from (98.35±10.12) to (30.35±7.31) (9<0.01),with significantly greater decline in amplitude and rate than that of the positive group (P<0.05).After treatment, the Amp of both groups showed a gradual increasing trend.Amp increased earlier in the positive group, but there was no significant difference in the extent of the increase between the two groups (P>0.05).After the treatment the CL and CMCT had shortened significantly in the negative group compared to before the treatment (P<0.05),while there was no significant change in CL and CMCT after the treatment (P>0.05).Conclusions The excitability of the contralateral motor cortex changes after a stroke.TMS can be used to characterize the MEP to monitor and predict recovery. This should help clinicians prepare more scientific rehabilitation plans.

ABSTRACT

Objective To research the clinical rehabilitative effect of repeated transcranial magnetic stimulation (rTMS) for the non-motor symptoms of Parkinson's disease. Methods Fifty-five PD patients were randomly divided into an rTMS treatment group (n =29) and a sham stimulation group (n =26).The treatment group received a course of 0.5 Hz rTMS treatment, while the sham stimulation group had the same treatment but with no energy output. Both groups were evaluated using a non-motor symptoms questionnaire (NMSQuest),the Hamilton depression scale (HAMD),the Pediatric Daytime Sleepiness Scale (PDSS),the mini-mental state examination (MMSE) and the scale for outcomes in PD for autonomic symptoms (SCOPA-AUT) before treatment, immediately after, and a month after treatment. Results Compared with before treatment, the average NMSQuest score of the treatment group declined significantly, though 1 month after treatment the improvement was no longer significant. After treatment and 1 month later the average NMSQuest score of the sham stimulation group increased gradually. The effect in the treatment group was therefore significantly better than in the sham stimulation group. After treatment and 1 month later the average HAMD score of the treatment group was significantly lower than before treatment while the average HAMD score of the sham stimulation group had increased gradually. The improvement in depression in the treatment group was therefore significantly better than in the sham stimulation group. After treatment, sleep disorders in the treatment group had been significantly alleviated, but one month later the improvement was no longer significant compared with before treatment.PDSS scores in the sham stimulation group declined gradually, but the improvement in sleep disorders among the treatment group was significantly better than among the sham stimulation group.Cognition improved significantly among the treatment group right after treatment, but 1 month later it had worsened while the MMSE scores of the sham stimulation group decreased gradually. The difference between the two groups was statistically significant. The average SCOPA-AUT scores of the two groups were not significantly different from each other or from the scores before treatment. Conclusion Repeated TMS can improve most non-motor symptoms of PD.The improvement in depression is the most significant.A short course of rTMS has no obvious rehabilitative effect on the autonomic function disorders of PD patients.

ABSTRACT

EEG characteristics in frequency domain were investigated in the frontal lobe, occipital lobe and hippocampus, i.e. cognition - related cortex of synaptic dysfunction model rat, providing electrophysiological basis for further study on plastic extent and nerve regeneration of the damaged neurons. Synaptic dysfunction model was made via microinjecting β - amyloid protein1 - 40 (Aβ1 - 40) into hippocampal CA1 area of rat. Morris water maze behavioral test was performed to evaluate the learning and memory function of model group. Then EEG in the above areas for two groups were recorded. The spectrum for two groups was performed and the characteristics in frequency domain were analyzed. The results showed: (1) The average escape latency in 3rd, 4th, 5th and 6th training times of model group are higher than those of normal. The average escape latency of normal group in 5th training time decreased more markedly than that in 2nd training time, while that of model group in 7th training time decreased more remarkably than that in 2nd training time (P <0.05). Without platform, the platform quadrant time percentage of model group was lower than the control (P <0.05). (2) Alpha rhythm in EEG of model rat was slowing down; alpha- band power decreased with peek frequency left shifted nearly 2Hz. And the power of delta - band and theta - band in frontal lobe, occipital and hippocampus all increased with different extent. The synaptic dysfunction model rats were made successfully by microinjecting Aβ1 - 40 method. The model rats show the decreased learning and memory dysfunction. EEG frequency spectrum features in model rat show slower alpha rhythm with power amplitude lower or loss, slow waves (delta and theta wave) increasing with higher power amplitude. These can be consistent with the EEG of Alzheimer's disease patients, which can provide electrophysiological basis for further plasticity and nerve regeneration study on the impaired cortex with synaptic dysfunction.

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the protective effects of vitamin B(12), ginseng saponin, and folic acid on mouse embryos subjected to high heat.</p><p><b>METHODS</b>Mice were used for the experiment.</p><p><b>RESULTS</b>After exposure of pregnant mice to high heat, the rates of teratism, stillbirth, and fetal absorption were markedly lower in mice treated with ginseng saponin and folic acid following heat exposure than in untreated mice. There were no significant differences in these rates when comparing mice treated with vitamin B(12) with the untreated mice.</p><p><b>CONCLUSIONS</b>Ginseng saponin and folic acid can lessen injuries to murine embryos caused by high heat, while vitamin B(12) has little protective effect against high temperature except for promoting overall embryonic growth.</p>
